摘要 目的探讨Stathmin/oncoprotein 18(Op18) mRNA和蛋白在肝细胞癌中的表达程度及其与肝癌临床病理特征的关系。方法应用RT-PCR技术检测33例肝癌、相应癌旁组织和肝癌细胞株SMMC7721和HepG2中Stathminm RNA的水平,同时应用免疫组织化学方法检测了上述组织和细胞株中Stathmin蛋白的表达。结果肝癌组织Stathmin mRNA的表达水平高于相应的癌旁组织(P<0.05)。肝癌组织Stathmin蛋白的阳性表达率为51.5%,癌旁组织为6%,阳性染色位于胞质内。两种肝癌细胞株中亦有Stathmin表达。Stathmin过表达与肿瘤大小、门脉受侵、TNM分期及Edmondson分级有关(P<0.05)。结论Stathmin在肝癌中过表达,可能与肝癌的发生及进展有关。 Objectives To study the expression of Stathmin/oncoprotein 18 (Op18) mRNA and protein in hepatocelluar carcinoma(HCC) tissues and to explore the relationship between the expression of Stathmin and clinical pathological features of HCC. Methods The expression of Stathmin mRNA in primary HCC (n = 33) and their correspond- ing adjacent tissues as well as hepatoma cell lines SMMC7721 and HepG2 were detected by semi-quantitative reversetranscription polymerase chain reaction (RT-PCR). The expression of Stathmin protein was observed in preceding cell lines and tissues by immunohistochemical staining. Results The expression level of Stathmin mRNA in HCC tissues was much higher than that in the matched surrounding non-cancerous tissues (P 〈0.05). Stathmin protein was 51.5% positive in HCC tissues and 6% positive in non-cancerous tissues and was mainly distributed in cytoplasm. The expression of Stathmin was also found in the two hepatoma cell lines. Stathmin overexpression was closely correlated with tumor size, portal vein invasion, TNM classification and Edmondson-Steiner' s classification (P 〈 0.05). Conclusions Overexpression of Stathmin may play an important role in development and progression of hepatocellular carcinoma.
