摘要
目的建立大鼠肾上腺嗜铬细胞癌PC12细胞2,5-己二酮(2,5-HD)中毒模型,观察研究神经生长因子(NGF)对2,5-HD中毒的保护作用。方法将细胞随机分为对照组(于正常培养基生长)、HD组(不同浓度2,5-HD)、NGF+HD组(不同浓度NGF+20mmol/L,2,5-HD),采用噻唑蓝(MTT)法检测细胞存活率。结果随着2,5-HD处理浓度升高和时间延长,细胞损伤加重,存活率较对照组逐渐降低(P<0.05,P<0.01);以NGF预处理的细胞显示出明显的抗损伤性,细胞存活率较HD组逐渐提高(P<0.05,P<0.01)。结论2,5-HD呈剂量性抑制PC12细胞存活和增殖,诱导细胞损伤和凋亡,NGF对2,5-HD中毒性PC12细胞显示出明显保护作用。
Objective: To set up a model of 2, 5 -HD intoxication of PC12 Cells of Rats'pheochromocytoma of adrenal; to observe and research the protective effect of nerve growth factor on 2, 5 - HD intoxication. Methods : Cells are divided by random into control group ( maintained in normal medium ) and HD group ( processed by 2, 5 - HD ) and NGF THD group processed by different concentration of NGF +20mmol/L, 2, 5 - HD) . MTT is adapted to detect the cells survivor rate. Resuhs: With the increasing of 2, 5 - HD concentration and the time extension, the cell injuries become worse and the sun, ivor rate decreases gradually (p 〈0. 05 ) ; the cells pre - processed by NGF show significant anti - injuring property and the survivor rate rises gradually (p 〈 0. 05), and NGF shows good protective effect on dose -dependent manner to protect PC 12 (P 〈 0. 001 ). Conclusions : 2, 5 - HD has the property of dose - dependent manner to inhabit the survival and hyperplasia of PC 12, induct cells injuries and withdrawing; NGF has obvious protective effect on 2, 5 - HD intoxication of PC 12 cells,
