氨基酮戊酸光动力作用诱导人宫颈癌Hela细胞凋亡的实验研究

摘要 目的:探讨光动力疗法(PDT)对体外培养的人宫颈癌细胞系Hela的杀伤效应。方法:将经不同浓度5-氨基酮戊酸(ALA)处理的Hela细胞照射不同剂量的激光,用MTT法测定OD492值,细胞染色观察形态学改变,流式细胞术检测细胞周期及凋亡率。结果:ALA10-1mM和激光剂量20J/cm2是PDT对Hela细胞杀伤作用的最佳参数。流式细胞术显示PDT组发生了G0-G1期生长停顿,凋亡率为(22.6±4.56)%,显著高于对照组。结论:ALA-PDT通过诱导凋亡对体外培养的Hela细胞产生杀伤作用。 Objective:To observe the effects of photodynamic therapy(PDT) on human cervical cancer cell line Hela in vitro.Methods:The cultured cells were treated with different concentrations of aminolevulinicacid(ALA) and different dosages of laser,then MTr colorometric assay was applied to measure the OD492.Cellular morphology was observe by May-Grtinwald-Farbstoff Giemsa staining.Cell cycle and apoptosis rate were estimated by flow cytometry.Results:The parameters of PDT is optimal when ALA was 10^-1mM and laser dosage was 20 J/cm^2.Flow cytometry showed that PDT group had GO/G1 stage arrest.The apoptosis rate in PDT group was (22.6+4.56)%.h was higher than that in control group.Conclusion:ALA-PDT has a significant killing effect on Hela cells by induction of apoptosis.
