摘要 以湿地松(PinuseliotiEngelm.)的未成熟合子胚为外植体,在附加8mg/L2,4D和4mg/LBA的LP培养基上诱导出胚性愈伤组织。在含1mg/L2,4D和05mg/LBA的培养基上保持并增殖。提高培养基的渗透压后,愈伤组织内形成大量的胚性胚柄细胞团和早期原胚。在附加4mg/LABA,75g/LPEG和5g/L活性炭的LP培养基上,早期原胚发育形成粗壮的子叶胚。在无激素LP培养基上,成熟的体细胞胚萌发并发育形成再生完整植株。体细胞胚转换成小植株的频率为156%。 Embryogenic callus was initiated from immature zygotic embryos of slash pine ( Pinus elliottii Engelm.) cultured on LP medium containing 8 mg/L 2,4 D and 4 mg/L BA. Multiplication can be achieved on LP medium with 1 mg/L 2,4 D and 0.5 mg/L BA. Embryonal suspensor masses and early stage proembryos were formed from embryogenic callus cultured on high osmotic pressure medium. Early stage embryos developed into cotyledonary embryos on LP medium supplemented with 4 mg/L ABA, 75 g/L PEG and 5 g/L activated charcoal. Mature somatic embryos germinated into regeneration plantlet on medium with hormone free. The frequency of somatic embryos converted into regeneration plantlets is 15.6%.
