摘要
目的:观察切除颌下腺对大鼠睾丸膜联蛋白5(annexin 5)表达的影响。方法:切除大鼠颌下腺,分别于术后14、28和42 d处死大鼠,Western blotting和免疫组织化学分析大鼠睾丸annexin 5表达及分布的改变。结果:Western blotting结果显示,与对照组相比,切除颌下腺14 d和28 d后,实验组大鼠睾丸annexin 5的表达分别升高了27.5%和35.2%,具有显著性差异(P<0.05,P<0.01);切除颌下腺42 d后,实验组大鼠睾丸annexin 5又恢复到与对照组几乎相同的水平。免疫组织化学显示,annexin 5在大鼠睾丸中的分布未见明显改变,但实验组annexin 5免疫组化显色较深,说明annexin 5的表达升高,这与Western blotting的结果基本一致。结论:颌下腺切除影响大鼠睾丸annexin 5的表达,颌下腺切除早期(14、28 d)颌下腺分泌的某些生物活性物质缺乏,而造成睾丸内annexin 5表达的升高。颌下腺切除后期(42 d),机体内因颌下腺切除而缺乏的某些物质在体内又重新得到了补偿,对睾丸annexin 5的影响消除,睾丸annexin 5的表达也恢复到正常水平。其内在具体机制尚待进一步阐明。
Objective : To investigate the expression of annexin 5 in rat testis after removing submandibular gland in rats. Methods:On day 14,28 and 42 after the operation, the changes of annexin 5 expression in rat testis were analyzed by Western blot and Immunohistochemistry. Results:Western blot showed that there was a 27.5% and a 35.2% significant increase (P 〈 0.05 ,P 〈0.01 )of the annexin 5 expression in rat testis on clay 14 and 28 respectively and there was no significant change in the expression of annexin 5 on clay 42 compared with that in control groups. Immunohistochemistry showed that the localization of annexin 5 in rat testis did not change on clay 14, 28 and 42 compared with that inthe control groups. Conclusion : Removing submandibular gland could affect the expression of annexin 5 in rat testis and this effect may be modulated by the hormones secreted from the submandibular gland, but the detail mechanism should be still explored.
