摘要
选取仙人掌多糖提取时间、提取温度和料液比3个因素进行二次回归正交组合设计试验,利用响应面法对其提取工艺参数进行优化。结果表明:在提取时间为132 min、温度为74℃、水体积为276 ml的条件下,仙人掌多糖提取产量最高,最大提取产量预测值为89.77μg/ml,实际提取值89.48μg/ml,二者基本相符。利用优化工艺参数提取仙人掌多糖时,具有最大的提取产量。
Effects of different temperature, time and the ratio of solid to solution on the extraction yield of polysaccharide from Opuntia were investigated based on composite design of quadratic regression. Extraction technique parameters were optimized with Response Surface Methodology. Experimental data were analyzed by solving the regression equation with MATLAB 7.0 software. It was indicated that the optimum extraction parameters were 74℃, 132 min and 276ml water volume. Under those conditions, the predicted value of polysaccharide extraction yield from Opuntia was 89.77 μg/ml, which was in consistent with the measured value of 89.48 μg/ml. This method with optimized technique parameters had maximal extraction yield of polysaccharide from Opuntia.
