摘要 目的:探索肠滤泡相关上皮细胞和M细胞的生物学特性。方法:取大鼠小肠PP结滤泡相关上皮细胞和M细胞进行培养,培养细胞分为对照组、内毒素组和灭活大肠杆菌组,分别予内毒素和灭活大肠杆菌干预,培养细胞爬片以抗细胞角蛋白-8抗体免疫荧光染色观察,培养液用ELISA检测TNF-α、IL-18、IL-27水平。结果:内毒素组TNF-α和IL-27水平升高,大肠杆菌组IL-18和IL-27升高,培养细胞部分呈抗细胞角蛋白-8抗体免疫荧光阳性。结论:肠滤泡相关上皮细胞和M细胞不仅在呈递抗原方面发挥作用,也可通过其分泌细胞因子,对机体局部和全身免疫功能产生影响。 Objective To study the biological characteristics of bowel M cell and follicle - associated epithelium of peyer' s patch in rats. Methods Epithelial tissue covering rat intestinal peyer' s patch was incubated and cultured. Bacterial endotoxin ( lipo - po- lysaochride, lμg/ml) or inactivated E. eoli ( 1 ×10^5/ml) was added into the specimens and furthur cultured for 6h. TNF -α, IL - 18, IL-27 contents of these culture fluid as well as the contents in the control specimens (nothing added) were measured with ELISA. Expressions of eytokeratin 8 (CK -8) in the culture cells were examined with inmuno -fluroseence assay. Rest.IllS The contents of TNF-α and IL - 27 in specimens cultured with adding LPS were significantly higher than those in control specimens ( P 〈 0. 05). The contents of IL - 18 and IL-27 in specimens cultured with adding inactivated E. coli were also significantly higher than those in control specimens (P 〈0.05). Anti - CK -8 antibody was demonstrated with immuno -fluroseence assay in a portion of culture cedis ( i. e. the M celia). Conclusion M cell and follicle associated epithelium of rat peyer' s patch could also secret cytokines to affect the local and general immune status of the animal.
