淫羊藿对肾阳虚雄性大鼠股骨骨形态发生蛋白-7表达的实验研究 被引量:7

摘要 目的观察淫羊藿对醋酸泼尼松所致的肾阳虚雄性SD大鼠股骨骨形态发生蛋白-7(BMP-7)表达的影响。方法3月龄雄性SD大鼠24只,随机分为正常组、肾阳虚模型组、淫羊藿组,灌胃给药90 d后,RT-PCR检测股骨BMP-7 mRNA的表达,免疫组化检测股骨BMP-7的表达。结果醋酸泼尼松致肾阳虚大鼠股骨BMP-7 mRNA和BMP-7的表达明显下降(P<0.05),淫羊藿能在mRNA水平(P<0.05)和蛋白水平上提高股骨BMP-7的表达。结论淫羊藿可提高股骨中BMP-7的表达来诱导成骨,修复骨损伤。 Objective To observe effects of Epimedium on the expression of bone morphogenetic protein-7 (BMP-7) in femoral bone of male rats with deficiency of kidney-Yang resulted from Acetic Prednisone. Methods Twenty four 3-month-old Sprague-Dawley male rats were randomly divided into three groups :normal group,deficiency of kidney-Yang model group and deficiency of kidney-Yang treated by Epimedium group,respectively. Rats had been oral gavages for ninety days,RT-PCR was applied to detect the expression of BMP-7 mRNA in femoral bone. Immunohistochemistry was applied to analyze the expression locations and quantities of BMP-7 in femoral bone. Results Prednisone reduced the expression of BMP-7 mRNA and BMP-7 in femoral bone significantly (P 〈 0. 05 ). Epimedium increased the expression of BMP-7 in femoral bone on the level of mRNA and protein ( P 〈 0. 05 ). Conclusion Epimedium increase the expression of BMP-7 in femoral bone to induce bone formation and rebuild injured bone.
