内皮祖细胞移植对糖尿病兔下肢缺血的病理及血管内皮生长因子的影响 被引量:7

Pathologic and VEGF changes of transplantation of endothelial progenitor cells for hindlimb ischemia in diabetic rabbit model

摘要 目的探讨内皮祖细胞(EPC)移植治疗糖尿病兔下肢缺血的病理及缺血肌肉中血管内皮生长因子(VEGF)的变化。方法2006年1月至9月在哈尔滨医科大学附属第二医院内分泌科将30只健康日本大耳白兔随机分为3组,即糖尿病磷酸盐缓冲液(PBS)对照组(A组)8只、糖尿病内皮祖细胞移植治疗组(B组)14只、正常血糖内皮祖细胞移植治疗组(C组)8只。骨髓来源的内皮祖细胞经体外扩增培养7d后,通过肌内注射进行细胞移植,用病理改变及肌浆中VEGF的变化评价治疗效果。结果EPC移植后14d,病理示3组毛细血管密度分别为(9.29±1.63)个/视野、(12.60±2.16)个/视野、(12.51±1.56)个/视野;血管数/肌束数分别为0.66±0.05,0.83±0.11,0.90±0.13;肌浆中VEGF质量分数[每克肌肉中VEGF因子的质量(ng)]分别为0.22±0.05,0.30±0.07,0.31±0.08;糖尿病内皮祖细胞移植治疗组与糖尿病PBS对照组比较,毛细血管密度、血管数/肌束数、VEGF值差异均有显著性意义,P<0.05。结论EPC移植能有效治疗糖尿病兔下肢缺血。 Objective To explore the effects of transplantation of endothelial progenitor cells (EPC) from autologous bone marrow for hindlimb ischemia in diabetic rabbit model, and the difference between diabetes and nondiabetes. Methods Rabbits were randomly allocated into three groups for transplantation therapy, PBS control group( n = 8, A group), EPC transplantation with diabetes group( n = 14, B group),EPC transplantation without diabetes group( n = 8, C group). Diabetic rabbit model was established by injecting alloxan, and hindlimb ischemia was induced by complete excision of femoral artery. Diabetic rabbits with hindlimb ischemia were therapied by transplanted ex vivo expanded EPC, and then effects were assessed by capillary density and capillary to nmscle fiber ratio, and VEGF in muscle was also determined. Results Capillary density,capillary to muscle fiber ratio,and VEGF in muscle increased significantly in EPC transplantation group with or without diabetes after transplanting 14 days ( P 〈 0. 05 ). Condnsion Endothelial progenitor cells transplantation is an effective therapeutic method for hindlimh ischemia in diabetic rabbit model.
