摘要
目的观察丹皮酚对脑缺血再灌注损伤一氧化氮(NO)和内皮素(ET)生成的影响。方法采用大脑中动脉内拴线阻断法(MCAO)制备大鼠局灶性脑缺血再灌注损伤模型,测定脑缺血1h再灌注12及24h后脑组织匀浆和血清中NO以及血浆中ET水平。结果大鼠脑缺血1h再灌注12及24h,脑组织匀浆和血清中的NO、ET水平均显著升高,丹皮酚(2.5,5,10mg·kg-1·d-1,po)给药7d,可显著降低脑缺血再灌注12和24h血浆ET及脑组织匀浆诱生型NOS(iNOS)活性和NO水平。结论丹皮酚减少脑缺血再灌注损伤时NO及ET生成,可能是其保护脑缺血损伤的作用机制之一。
OBJECTIVE To investigate the effects of paeonol on nitric oxide and endothelin production in focal brain ischemia-reperfusion injury rats. METHODS The NO and ET levels of brain homogenate,serum and plasma were measured with a model of reversible middle cerebral artery occlusion(MCAO) without craniectomy in rats. RESULTS The levels of NO and ET were increased significantly in ischemia-reperfusion rats compared with sham rats. Paeonol decreases the activity of iNOS,levels of NO in brain homogenate and decreases the plasma levels of NO and ET in 12 and 24 h following reperfusion compared with ischemia-reperfusion injury rats. CONCLUSION The results suggested that the effect of paeonol decreasing NO and ET production may play an role on amelioration of focal brain ischemia reperfusion injury.
