通络Ⅳ号对局灶性脑缺血再灌注损伤大鼠神经细胞凋亡的影响 被引量:4

摘要 目的:探讨中药复方通络Ⅳ号对脑缺血再灌注模型大鼠脑神经细胞凋亡的影响。方法:成年雄性Wistar大鼠30只随机分为假手术组、治疗组、对照组3组,用线栓法制备气虚血瘀证大鼠大脑中动脑闭塞(MCAO)脑缺血再灌注(IR)模型,参照Zea-Longa法进行神经病学评分、TUNEL法检测凋亡细胞。结果:假手术组神经功能缺失评分为0分,仅见极少量凋亡细胞。治疗组的神经功能缺失评分为1.80±1.03分,显著低于对照组的2.90±1.10分(0.01<P<0.05);治疗组凋亡细胞计数为79.90±34.92个/层,对照组凋亡细胞数为145.50±50.24个/层,两组均数经变量变换后进行比较存在显著差异(P<0.01)。通络Ⅳ号治疗组的神经病学评分与凋亡细胞数均明显低于生理盐水对照组。结论:复方中药制剂通络Ⅳ号能明显减轻脑缺血再灌注动物的神经功能缺损与脑神经细胞凋亡。 Objective : To explore the effects of Tongluosihao on apoptosis in brain tissue of ischemia reperfusion (IR) model Rats. Methods: Middle cerebral artery occlusion (MACO) was taken to establish the IR model rats. Longa scores and apoptosis cells examined by TUNEL staining were determined and used to evaluate the effetcs of Tongluosihao on neural dysfunction and apoptosis in brain tissue of IR model rats. Results: Longa scores in the treatment gruop ( 1.80 ± 1.03) were significantly lower than those in the control group (2.90 ± 1.10) (0. 01 〈P 〈0. 05). Apoptosis cells in the treatment gruop (79.90±34.92) were significantly lower than those in the control gruop ( 145.50 ± 50. 24) (P 〈 0. 01 ). Conclusion : Tongluosihao can reduce the neural dysfunction and apoptosis in brain tissue of IR model Rats.
