摘要
目的探讨副粘病毒Tianjin株的种系进化地位。方法将副粘病毒Tianjin株HN核苷酸、氨基酸序列与GenBank发布的副粘病毒相应序列进行比较,构建种系进化树,阐明副粘病毒Tianjin株在副粘病毒科中的分类地位。结果基于HN核苷酸序列的种系进化分析表明,副粘病毒Tianjin株属于副粘病毒科、副粘病毒亚科、呼吸道病毒属,与仙台病毒亲缘关系最近。Tianjin株与仙台病毒BB1株HN核苷酸、氨基酸序列同源性分别为94.8%、95.1%,与其他仙台病毒,在84.8%~88.7%和89.6%~91.7%之间。Tianjin株与BB1株HN核苷酸序列的差异明显大于同一进化分支内的仙台病毒之间的差异。Tianjin株HN蛋白存在18个独特的氨基酸残基变异。结论结合种系进化分析结果、病毒的宿主来源和致病特点,提示副粘病毒Tianjin株不属于仙台病毒现有的3个进化分支而自成独立的一支。
Objective To explore the phylogenetic location of paramyxovirus Tianjin strain. Methods The homologous and phylngenetic analysis of hemagglutinin-neuraminidase (HN), or hemagglutinin (H), or G nucleotide sequences and deduced amino acid sequences between the paramyxovirus Tianjin strain and viruses in the seven genera of paramyxoviridae family from GenBank were performed. The phylogenetic tree was constructed. Results Tianjin strain is a member of the Respimvirus genus in the Paramyxoviridae subfamily, Paramyxoviridae family and has a close relationship with Sendai virus. The homology of HN nucleotide and amino acid sequences between paramyxovirus Tianjin strain and Sendai virus BB1 strain are 94.8%, 95.1%, respectively, whereas the identity are 84.8%-88.7% and 89.6%-91.7% between Tianjin strain and various Sendai viruses. The divergence of DIN nucleotide sequences between Tianjin strain and BB1 strain is much more evident than that of Sendai viruses within the either evolution lineages, one is represented by Sendai virus Hamamatsu strain and Ohita strain, the other includes Fushimi strain, Nagoya strain, pi strain and Z strain. There are 18 unique amino acid substitutions in the HN protein of Tianjin strain. Conclusion Paramyxovirus Tianjin strain doesn't belong to any of the three known evolution lineages of Sendai virus.
