摘要 将体外培养的小鼠囊胚分别施以38℃温和热应激和40℃强烈热应激处理1、2和3 h,然后在37℃正常培养条件下分别恢复3、2和1 h,用体视显微镜观察囊胚孵出率以及经38℃和40℃诱导热应激后囊胚的孵出率,用Western-blot技术检测小鼠囊胚HSP72的表达。结果,38℃热应激处理2 h时,HSP72表达量达到最高水平,与对照组差异显著;40℃热应激处理3 h时,HSP72表达量达到最高水平,与对照组差异不显著;38℃热诱导2 h囊胚孵出率最高,与40℃强烈热应激处理2 h组差异显著。结果表明,热诱导组囊胚孵出率与38℃热应激组囊胚HSP72的表达量呈正相关。 Mice embryos cultured in vitro to blastocysts were exposed to either 38 ℃ mild heat stress or 40 ℃ severe heat stress for 1 h,2 h and 3 h,respectively,and followed by cultivation at 37 ℃ for 3 h,2 h and 1 h,respectively. Stereomicroscope was used to examine the hatching rate of the blastocysts and the hatching rate of the blastocysts underwent induced heat stress by cultivation at 38℃ and 40 ℃, respectively. Western-blot was performed to determine the expression of HSP72 in the heat stressed blastocysts. The expression of HSP72 reached the highest level in the embryos stressed at 38 ℃ for 2 h and it was significantly higher than that in the control group. The expression of HSP72 reached the highest level after the embryos were treated at 40 ℃ for 3 h and it was not significantly different from that in the control group. The hatching rate reached the highest level when the embryos were treated at 38 ℃ for 2 h and it was significantly higher than that in the embryos treated at 40 ℃ for 2 h. The results indicated that the hatching rate of the blastocysts in thermal induction group had the positive correlation with the expression of HSP72 in blastocysts treated at 38 ℃.
