摘要 为了建立小白菜线粒体DNA(mtDNA)提取体系,以小白菜黄化苗为材料,研究了不同提取方法、材料选择等因素对线粒体DNA提取的影响。结果表明,经冰浴研磨小白菜黄化苗,通过简易的密度梯度离心技术快速抽提大量高纯度的线粒体,蛋白酶K裂解线粒体及有机溶剂抽提去除蛋白质,可获得纯度较高的mtDNA,能用于RAPD分析。 In order to establish the extraction method of Chinese cabbage mitochondrial DNA (mtDNA),the effects of the different methods,material selection on the mtDNA extraction from etiolated seedling of Chinese cabbage were inrestigated.The results indicated that after grounding the etiolated seedlings on ice and,extracting mitochondria by simple density gradient centrifugation, and then the mitochondria was ruptured by the proteinase K and organic solvent,high purity mtDNA were isolated,which could be used for the RAPD analysis. It was a simpler, faster and more economic method than the general methods.
