昆明鼠腹腔注射甲苯急性中毒的行为学实验研究

A Study of Acute Toxicity of Toluene on the Behavior of Kunming Mice

摘要 目的研究昆明鼠腹腔注射甲苯急性中毒的行为学表现,探讨甲苯急性毒性神经损伤。方法昆明鼠随机分成6组,8 000mg/kg,4 000mg/kg,2 000mg/kg,1 000mg/kg,500 mg/kg等不同剂量甲苯小鼠腹腔注射制作急性中毒模型,生理盐水对照组,染毒后观察24h一般行为学改变和4d、6d、8d、10d、12d、14d运用程控自主活动箱检测神经兴奋性变化,运用寇氏法公式计算药物半数致死剂量LD50。结果甲苯染毒各组都有急性中毒的表现,主要表现为抑制;甲苯急性毒性对小鼠有严重损伤,其自主活动表现为短暂的兴奋后迅速转为抑制。LD50=1782.38?/?,S lg LD50=0.0602(LD50的标准误),LD5095%C I(可信区间)=(1358.31 2338.84)?/?。自主活动明显降低(P<0.05),2 000 mg/kg与1 000 mg/kg、500 mg/kg比较有差异(P<0.05)。结论小鼠腹腔注射甲苯发现具有急性毒性,LD50=1782.38?/?;中毒小鼠自主活动性降低(P<0.05)。 Objective To observe the behavioral responces of Kunming mice after intra - peritoneal injection of different doses of toluene. Methods The mice were divided into different groups 18000mg/kg,4000mg/kg,2000mg/kg, 1000rag/ kg,500 mg/kg). The influences of toluene acute toxicity on mice autonomic activities in 24h could also be known. All these operations were carded out in 4d, 6d, 8d, 12d, 14d respectively. We established intra - peritoneal injection models of toluene and observed ethology, nervous excitation and LD50. Results Toluene had acute toxicity. The mice groups using narcotics all showed acute toxic performances such as: inhibition activities, writhing, dull reactions, still bending, squinting eyes, the ocular region secreting, sweating, the breathlessness turning into asthenia, appetite degrading and the death. To use Karber' s method LD 5o = "1782.38"mg/k9, Slg LD50 = "0. 0602", LD5095% CI = " ( 1358.31 2338.84 ) "mg/kg. The obvious deduction of the automatic activities number ( P 〈 0.05 ) , which indicated there were differences available among 2000 mg/kg and 1000 mg/kg, 500 mg/kg ( P 〈 0.05 ). Conclusion The toxic effects of the toluene on the mice were found after intra - peritoneal injection, LD 50 = 1782.38 mg/kg. The mice automatic activities were decreased ( P 〈0.05 ).
