摘要
目的研究脑胶质瘤中细胞周期素D1(cyclin D1)表达与胶质瘸细胞恶性程度及细胞增殖活性的关系。方法采用免疫组化方法检测46例脑胶质瘤和7例正常脑组织标本中cyclin D1和增殖细胞核抗原(PCNA)的表达水平,观察并分析各病理等级中两者表达的阳性率和标记指数。结果脑胶质瘤组cy- clin D1蛋白阳性表达率60.9%(28/46),显著高于正常脑组织组的14.3%(1/7)(P<0.05)。随着人脑胶质瘤病理分级的增高,cyclin D1的阳性率明显升高,在高、低恶性度组间差异具有统计学意义(64.2% vs 45.5%,P<0.05)。经Spearman相关性分析发现cyclin D1与PCNA的平均标记指数呈明显正相关(r=0.576,P<0.05)。结论人脑胶质瘤中cyclin D1的表达与细胞增殖活性和病理学分级关系密切,可能对肿瘤细胞的增殖和恶性转化产生重要的推动作用。
Objective To study the expression of cyclin D1 in human ghomas and its relationship with pathological grades and cellular proliferation activity. Methods The expression of cyclin D1 and proliferating cell nuclear antigen (PCNA) were detected by immunohistochemistry in 46 human brain gliomas and 7 normal brain tis- sues to observe their positive rates and the labcling indices in each WHO grade of human brain gliomas. Results In human brain gliomas,the positive rates of cyclinD1 expression (60.9%) was remarkably increased compared with matched normal tissues ( 14.3% ) ( P 〈 0.05 ). Cyclin D1 positive rates increased with the advancing pathological grades. Significant difference was seen in positive rates between low grade ( Ⅰ + Ⅱ ) and high grade ( Ⅲ+ Ⅳ ) (45. 5%vs 64, 2% ) (P 〈0.05 ), Positive correlation was obtained between the labeling indices of cyclin D1 and PCNA by Spearman correlation analysis ( r = 0. 576, P 〈 0.05 ). Conclusion Expression of cyclin D 1 is closely correlated with the cellular proliferation activity as well as pathological grades in human brain gliomas. It may he an important factor in promoting cell proliferation and transformation to malignant phenotype.
