摘要
针对以苯甲醛、氯仿为原料合成苯甘氨酸的反应过程,建立了高效液相色谱法测定苯甘氨酸含量的方法,考察了流动相组成与流量对分离效果的影响,得出了适宜的分析条件。以10%甲醇+90%磷酸二氢钠溶液(20 mmol/L)为流动相,在其流量1 mL/min、柱温为室温和检测波长260 nm的条件下,苯甘氨酸质量浓度在25~400 mg/L范围内,质量浓度与峰面积呈较好的线性关系,系统精密度指标RSD值为0.13%。
The high performance liquid chromatography(HPLC) method for the analysis of phenylglycine in the synthesis of phenylglycine from benzaldehyde and chloroform was established. The influences of the composition and the flow-rate of the mobile phase on the separation were examined. The optimum chromatography conditions were found as follows: V(methanol) : V[monosodium phosphate (20 mmol/L)]=1: 9 as the mobile phase, flow-rate of mobile phase 1 mL/min, detection wavelength 260 nm. The standard curve for phenylglycine was linear over the range of 25 -400 mg/L. The RSD value for the accuracy of the system was 0. 13%.
