大鼠重组EPO结合蛋白的表达纯化及活性鉴定
摘要
EPO是调控骨髓红系造血的关键因子,其与红系祖细胞表面的特异受体EPO—R结合引发胞内信号机制,防止细胞凋亡并促进其增殖和分化。EPO—R胞外域是与EPO发生结合的受体区段,又称为EPO结合蛋白(EBP)。迄今,采用基因工程方法制备的EBP主要被应用于受体一配体结合特性及物理结构研究,其在治疗EPO反应性增生失调如红白血病和红细胞增多症中的可能性探讨尚未见报道。我们依据EPO-R与EPO结合的化学计量特性设计了串联融合的双体结合蛋白,采用原核系统进行表达,对其体内外结合活性进行了鉴定。
