激光照射联合龙葵多糖对荷瘤小鼠肿瘤细胞凋亡的影响 被引量:4

摘要 目的:探讨低能量激光照射联合复合多糖对荷肝癌小鼠肿瘤细胞周期及凋亡的影响。方法:建立荷肝癌小鼠皮下种植瘤模型,将荷瘤小鼠随机分为5组,分别为NS组、低能量激光照射组、多糖组、低能量激光照射联合多糖组及5-氟尿嘧啶(Fl’uorouracilum,5-Fu)组。对照组给生理盐水灌胃,治疗组给复合多糖、5-Fu灌胃,用低能量激光照射脾区进行治疗,连续治疗10天后将小鼠处死。取肿瘤组织用流式细胞仪检测细胞周期及细胞凋亡;免疫组化法检测瘤组织中凋亡抑制因子Survivin蛋白的表达情况。结果:低能量激光照射联合多糖可以使肿瘤细胞停止于G1期,促进肿瘤细胞凋亡,并显著下调Survivin蛋白的表达。结论:低能量激光照射联合多糖能够调控肿瘤细胞周期、诱导肿瘤细胞凋亡,抑制荷肝癌小鼠肿瘤的生长。 Objective To study effects of low energy laser irradiation unite Herba Solani Nigri Polysaccarides on tumor cell cycle and cell apoptosis in liver cancer loaded mouseMethod Established liver cancer loaded mouse model and divided randomly roice intofive groups, namely, NS groups, lowj energy laser irradiation group, Herba Solani Nigri Polysaccharide group, low energy laser irradiation unite Herba Solani Nigri Plysaccharide group and 5' FU group. NS was given to the control group orally, Herba Solani Nigri Polysaccharide and 5 - FU was given to the treated group in the same way, and use the low energy laser irradiation to light up the spleen every day. Then executed mice after 1 0 days and took tumor issue to measure cell cycle distribution and cell apoptosis, 析 m flow cytometry. We detected the expression of Survivin protein/with immune histochemistry. Result low energy laser irradiation unite Polysaccharide could arrest tumor cell at G1 stage, induce tumor cells into apoptosis, and the expression of Survivin protein wele decreased. Conclusion low energy laser irradiation unite Polysac charide Can regulate the tumor cell cycle,induce tumor cells into apoptosis and inhibit the growth of the liver cancer in mice.
