摘要
目的观测银杏苦内酯B(BN52021)干预前后ANP大鼠肺组织血小板活化因子受体(PAF-R)mRNA和蛋白质表达的变化,探讨其作用机制。方法180只大鼠随机分为对照组(NC组)、ANP组和BN52021治疗组(BN组),每组再分为制模后1h、2h、3h、6h、12h、24h6个亚组。逆行胰胆管内注射牛磺胆酸钠诱导ANP。BN组于制模后15min股静脉注入BN52021(0.5mg/100g体重)。各时间点取血检测血清淀粉酶,取胰腺和肺组织行病理学检查,采用RT-PCR和Western blot检测肺组织PAF-R mRNA和蛋白质表达。结果ANP组和BN组各时点血清淀粉酶均高于NC组(P〈0.05),BN组1h、2h、3h、6h、24h时点均低于ANP组(P〈0.05)。ANP组和BN组胰腺和肺的病理分值均高于NC组,BN组在3h、6h、12h显著低于ANP组(P〈0.05)。ANP组和BN组PAF-R mRNA和蛋白质水平分别在3h和1h显著高于NC组,BN组与ANP组相比无显著差异。结论PAF-R在ANP早期急性肺损伤中起重要作用,BN52021对其表达无显著影响。
Objective To investigate the role of PAF-R in the pathogenesis of acute lung injury (ALI) of acute necrotizing pancreatitis (ANP) in rats and the effect of Ginkgolide B (BN52021) on expression of PAF-R in pulmonary tissue. Methods 180 male Wistar rats was randomly allocated to three groups: normal control group (NC, n = 60), ANP model group (ANP, n = 60) and BN52021 treatment group (BN, n = 60). ANP were induced by retrograde injection of sodium taurocholate into the biliary duct. Each group was further divided into six subgroups according to different time-points after induction (1, 2, 3, 6, 12 and 24 h) (n = 10). The rats were sacrificed at the respective time points, then the blood was collected to evaluate the levels of serum amylase, and pancreas and lungs tissues were obtained to be studied pathologically. The levels of PAF-R mRNA and its proteins expression in lung tissues of rats were detected by RT-PCR and Western blot, respectively. Results The levels of serum amylase and pathological changes in pancreas indicated that ANP model was induced successfully. The levels of serum amylase in ANP and BN group were significantly higher than those in NC group at different time-points (1, 2, 3, 6, 12 and 24 h) (P〈 0.05), while the levels of serum amylase in ANP group were significantly higher than those in BN group at 1, 2, 3, 6, 24 h (P 〈 0.05). The scores of pathological changes of ANP and BN group were significantly higher than those in NC group, while the scores of pathological changes of ANP group were significantly higher than those in BN group at 3, 6, 12 h (P 〈 0.05). The levels of PAF-R mRNA and proteins expression in lung tissue in ANP and BN groups were significantly higher than those in NC groups at 3h and lh (P〈 0.05), respectively. The levels in BN groups were not significantly different from those in ANP groups. Conclusions PAF-R played an important role in early phase of ALI complicated with ANP. BN52021 had no significant effect on the expression of PAF-R.
