摘要
目的探讨基质金属蛋白酶-9(MMP-9)、基质金属蛋白酶抑制剂-1(TIMP-1)和血管内皮生长因子(VEGF)在视网膜母细胞瘤(RB)中的表达及其与RB分化程度和视神经浸润的关系。方法应用MMP-9、TIMP-1单克隆抗体、VEGF多克隆抗体,采用PV6000免疫组化方法,对32例RB常规石蜡包埋标本进行MMP-9、TIMP-1和VEGF表达的测定。结果MMP-9和VEGF在RB中表达阳性率分别为56.25%(18/32)和68.75%(22/32),均与RB的分化程度及视神经浸润有关(P均<0.05)。TIMP-1的阳性表达率(18.75%,6/32)与RB的分化程度及视神经浸润无关(P>0.05)。MMP-9和VEGF在RB中的表达明显正相关(P<0.05)。结论MMP-9可以通过激活VEGF并上调VEGF的表达,促进RB组织中新生血管的生成。
[ Objective] To investigate the expression of matrixmetalloproteinase-9 (MMP-9), tissue inhibitor of metal- loproteinases-1 ( TIMP-1 ) and vascular endothelial growth factor (VEGF) in retinoblastoma (RB) and the relationship with the differentiation and optic nerve infiltration. [ Methods] The expression of MMP-9, TIMP-1 and VEGF was examined in 32 cases of RB tissue by using PV6000 immunohistochemistry. [ Results ] The positive rates of MMP-9 and VEGF were 56. 25% and 68.75 % respectively, and all the expression were significantly related to the differentiation degree and nerve infiltration (P 〈 0. 05 ). The positive rate of TIMP-1 expression was 18.75%, and not correlated with the differentiation degree and nerve infiltration (P 〉 0. 05). The expression of MMP-9 was positively correlated with the expression of VEGF in RB (P 〈0.05). [Conclusions] MMP-9 may activate VEGF, and promote the tumor angiogenesis of RB by up-regulating the expression of VEGF.
