SPE-HPLC测定断血流片中醉鱼草皂苷Ⅳb的含量

Determination of the content of buddlejasaponins Ⅳb in duanxueliu tablet by HPLC

目的:建立固相萃取-高效液相色谱法测定断血流片中醉鱼草皂苷Ⅳb的含量。方法:色谱柱Dikma Kromasil C18柱(5μm,250mm×4.6mm),流动相甲醇–水(80∶20),流速0.8ml/min,检测波长250nm,柱温40℃。结果:醉鱼草皂苷Ⅳb在0.7715～6.136μg范围内具有良好的线性关系(r=0.9999),平均回收率为98.57%,RSD为0.56%(n=9)。结论:该方法简便、快速、有效、灵敏、准确,具有良好的重复性和回收率,可作为该制剂的定量分析方法。

Objective ：To establish a method for the content determination of buddlejasaponins 1Vb in duanxueliu tablet by solid phase extraction and HPLC. Methods ：The separation was carried out on a Dikma Kromasil C18 column（5μm ,250 mm×4.6 mm）;The mobile phase was methanol-water（80：20）;The flow rate was 0.8 ml/min ;The detective wavelength was set at 250 nm ;The column temperature was 40 ℃.Results：The calibration curve showed good linearity over the range of 0.7715-6.1361μg （r=0.9999）. The average recovery was 98.57% with RSD 0.56% （n=9）.Conclusion ：This method was simple, rapid ,efficient, sensitive and accurate,with good repeatability and recovery,it could be used as a quantitative analysis method for this preparation.
