摘要
目的:观察针刺对慢性全脑缺血缺氧性大鼠认知功能、血浆中血栓素B2(TXB2)和6-酮-前列环素(6-酮-PGF 1a)的影响。方法:30只健康雄性Wister大鼠随机分为针灸组,对照组,模型组各10只。针刺组和模型组采用Pul-simelli法制备大鼠全脑缺血模型,并配合行为学测试确立模型成功。术后24 h,针刺组开始针刺大鼠百会、四神聪、涌泉,内关、水沟、合谷、三阴交、足三里、太冲穴,每周5次,每次10 min;模型组则给予腹腔注射0.9%生理盐水(2 ml/kg)。术后24 h、5及21 d时进行大鼠神经行为学评估、检测血浆中TXB2和6-酮-PGF 1a的水平,并在第21天应用Y迷宫评价大鼠的认知功能。结果:针刺组大鼠在针刺治疗后认知功能测试成绩和神经行为学评分均明显高于模型组(P<0.01),血浆中TXB2的水平显著下降而6-酮-PGF 1a的水平略有上升。结论:针刺对慢性全脑缺血缺氧性大鼠的认知功能有显著的改善作用,对神经行为学的损伤具有较明显的修复作用。
Objective:To observe the mechanism of acupuncture on the cognition function and the contents of serum thromboxan B2 and prostacyclin in rats with chronic total cerebral ischemia-hypoxia. Methods: Thirty male Wistar rats were randomly divided into the acupuncture group, the control group and the model group (n= 10 in each group). In acupuncture group and model group, the Pulsimelli method was used to prepare total cerebral ischemia model in rats. Twenty-four hours after operation, the points of Baihui, Sishencong, Yongquan, Neiguan, Shuigou, Hegu, Sanyinjiao, Zusanli and Taichong were acupunctured in acupuncture group, three times every week, 15 min/time. The rats in the model group were intraperitoneally injected with 0.9% normal saline (2 ml/kg). At 24th hours, 5th day and 21st day after operation, neurobehavioral functions in rats were evaluated and the contents of serum thromboxan B2 and prostacyclin determined. At 21st day, cognition function of the rats was assessed by Y maze. Results: After acupuncture in the rats with chronic total cerebral ischemia-hypoxia, the performance record of cognition function and neuroethology score were elevated obviously (P〈0.01), and the contents of serum thromboxan B2 were decreased but the contents of prostacyclin raised slightly. Conclusion: Acupuncture could improve the cognition function and restore nervous detriment of rats with chronic total cerebral ischemia-hypoxia.
