摘要
目的探讨胃黏膜病变中幽门螺杆菌(Hp)感染与环氧化酶-2(COX-2)、核转录因子(NF-κB)表达的关系。方法应用免疫组织化学染色方法(S-P法)分别检测COX-2、NF-κB蛋白在人慢性浅表性胃炎、慢性萎缩性胃炎、不典型增生及胃癌组织中的表达情况。应用Giemsa染色方法及快速尿素酶试验判定Hp感染。结果慢性萎缩性胃炎、不典型增生、胃癌各组Hp阳性者COX-2及NF-κB的阳性表达显著高于Hp阴性者,Hp阳性各组间COX-2和NF-κB的阳性率呈逐渐升高趋势。Hp阴性各组间COX-2和的NF-κB表达无显著性差异。COX-2与NF-κB在胃黏膜病变中的表达具有显著相关性。结论在胃黏膜病变中COX-2与NF-κB的表达与Hp感染有关,介导COX-2与NF-κB表达可能是Hp感染致癌的重要机制之一。
Objective To study the relationship between the expression of cyclooxygenase-2(COX-2) and nuclear factor-kappa B (NF-κB) protein and H.pylori infection. Methods COX-2 and NF-κB protein expression were examined by immunohistochemistry. H.pylori infect was examined by Giemsa and urease tests. Results Positive rate of NF-κB and COX-2 expression in gastric carcinoma group, atrophic gastritis group and chronic atrophic gastritis with positive H.pylori was higher than these groups with negative H.pylori. The positive rate of NF-κB and COX-2 expression in H.pylori positive groups became higher and higher from chronic superficial gastritis to gastric carcinoma, but there was no significant difference between H.pylori negativeand positive group. The expression of COX-2 and NF-κB protein was closely related to gastric mucosal lesions. Conclusions We suggest that NF-κB and COX-2 may be associated with H.pylori infection in gastric mucosal lesions. Expression of NF-κB and COX-2 caused by H.pylori infection may be one of the important mechanisms of gastric carcinoma.
