摘要
通过比较两种肽类激素(激活素和抑制素)对细胞增殖与激素分泌的作用,了解它们作用方式的差异。以来源于人胎盘的绒毛膜上皮癌细胞系Jar细胞为实验材料,利用MTT法测定细胞增殖,利用放射免疫法测定培养液中的孕酮含量,研究了激活素A和抑制素A对Jar细胞的增殖和孕酮分泌的影响。结果表明,激活素A(1~100μg/L)对细胞增殖有明显的促进作用,且呈现浓度依赖性,浓度为30μg/L时作用最强。与之相对比,抑制素A对Jar细胞的增殖无显著作用。同时,激活素A能刺激Jar细胞分泌孕酮,最大作用浓度为10μg/L。抑制素A对Jar细胞分泌孕酮则没有明显的影响,以上结果提示,激活素A对人绒毛膜上皮癌细胞的增殖和激素分泌起着较明显的调节作用,而抑制素A则未见直接作用。
Cell proliferation and progesterone content in culture medium were determined by MTT test and by radioimmunoassay respectively.Activin A could stimulate cell proliferation and the effect was in a dose dependent manner,while the concentration of 30μg/L was most effective.In contrast,inhibin A had no obvious effect on cell proliferation.Activin A could also stimulate the progesterone production,but inhibin A could not.The most effective concentration of activin A was 10μg/L for progesterone stimulation.The results indicate that activin A may play an important role in regulating the proliferation and progesterone secretion of human choriocarcinoma cell line,but the role of inhibin A is still obscure.
