摘要 目的:观察四物汤是否对实验性血栓形成具有抑制作用,并初步探讨其机制。方法:家兔随机分为盐水对照组与四物汤组。用颈外静脉结扎、颈总动脉双氧水损伤和凝血酶注射法分别制成静脉内、动脉内血栓模型和血液高凝状态,然后测定血栓的长度、重量和血浆部分反应指标的变化。结果:盐水对照组体外形成的血栓长13.4±4.0cm,干重为66.0±20.0mg,静脉内血栓长15.6±2.9cm,干重40.1±9.7mg;动脉内血栓四物汤组为0.53±0.30mg/mg血管重,盐水对照组为1.48±0.15mg/mg血管重。四物汤组各数值均明显低于盐水对照组(P均<0.01)。凝血酶组部分凝血活酶时间(APTT)、凝血酶原时间(PT)和凝血酶时间(TT)均较盐水对照组明显缩短(P<0.05、P<0.05和P<0.01);而四物汤则能使之延长,延长率分为18%、19%和39%,P<0.01。四物汤组血管性假血友病因子(vWF)为82.2%±7.5%,前列腺素B2(TXB2)为65.6±26.1ng/L,明显低于凝血酶组;而6酮前列腺素F1α(6ketoPGF1α)为38.1±7.2ng/L,明显高于凝血酶组;组织型纤溶酶原激活因子(tPA? Objective:To determine the inhibitory effects of “si wu tang” on experimental thrombosis and its mechanism.Methods:Rabbits were at random divided into the control and “si wu tang” groups.Experimental thrombosis was induced by ligation of external jugular vein and injection of H 2O 2 solution in arteria carotis communis.The thrombi were taken out and the length and dry weight were determined.Another ten rabbits were divided into thrombin and thrombin plus “si wu tang” groups,thrombin was intravenously injected following the injection of “si wu tang” or normal saline.three hr later,blood samples were drawn.APTT,PT and TT were measured.The concentrations of tPA and vWF in plasma were determined by ELISA,and TXB 2,6ketoPGF 1α in plasma and GMP140 on platelet by RIA.Results:The thrombi of “si wu tang” group formed in vitro were 2 7±7 5mg and 15 6±6 0mg in vivo.In contrast with that of the controls(66 0±20 0mg,40 1±9 7mg),“si wu tang” remarka bly inhibited thrombosis( P <0 01).The mean values of APTT,PT and TT of the samples from thrombin plus “si wu tang” were 46 2±1 3second,16 6±1 1second,and 2 2±1 9 second respectively.Compared with thrombin group,these changes were significantly prolonged( P <0 01).The concentrations of TXB 2,vWF and GMP140 in thrombin group were 127 6±20 1ng/L,183 4%±22 4% and 1845±536 n/BPC, which were markedly higher than that of thrombin plus “si wu tang” group( P <0 05).However, 6ketoPGF 1α (38 1±7 5ng/L) in thrombin plus“si wu tang” group were remarkably increased than in thrombin group,and tPA in thrombin plus “si wu tang” group(9 6±0 9μg/L)were remarkably increased than in controls( P <0 05).Conclusions:“si wu tang” has antithrombotic effects and it is associated with the inhibition of blood coagulation and platelet activation and the promotion of ability of vascular endothelial cells to antithrombosis.
