肝癌组织TGF-β1和IGF-II表达水平与HBV复制关系 被引量:1

摘要 目的:探讨肝癌(HCC)组织HBV复制与转化生长因子TGF-β1和胰岛素样生长因子IGF-II表达间的关系。方法:以自身对照法收集人HCC组织及非癌组织,以生物素标记的HBV-DNA探针检测HCC组织中HBV-DNA;采用免疫组织化学方法,检测组织中TGF-β1和IGF-II的表达;分析TGF-β1和IGF-II表达与HBV复制间的临床病理学关系。结果:HCC组织中TGF-β1和IGF-II均呈较高表达,在HCC癌灶及非癌组表达存在显著差别,其阳性率均为83.3%,HCC组明显高于非癌组(P<0.01)。HCC的癌灶组中TGF-β1和IGF-II阳性表达与肿瘤分化程度显著相关(P<0.05);而与肿瘤直径、数目无关(P>0.05);TGF-β1和IGF-II表达与HBV复制显著相关,且HBVDNA阳性组显著高于HBV-DNA阴性组。结论:肝癌组织中TGF-β1和IGF-II过度表达,且与HBV复制和肝癌的分化程度有关。 Ojective :To investigate the relationship between hepatitis B Virus (HBV) replication and the expressions of transforming growth factor (TGF)-β1 and insulin-like growth factor-Ⅱ (IGF-Ⅱ) in human hepatocellular carcinoma (HCC). Methods:Liver HBV-DNA was detected by in situ molecular hybridization technique, and the expressions of TGF-β1 and IGF-Ⅱ were detected by the immanohistochemistry in HCCs and their self-control non-cancerous tissues. The relationship was investigated between TGF-β1 or IGF-Ⅱ expression and HBV replication or their clinical pathological characteristics. Results :The stronger expressions (83.3%) of TGF-β1 and IGF-Ⅱ were found in HCC tissues. A significant difference was presented between in HCC tissue and in non-cancerous liver tissues (P〈0.01). The positive expression of TGF-β1 in HCC was correlated to tumor differentiation, but neither to tumor size nor numbers (P〉0.05). The levels of TGF-β1 and IGF-Ⅱ expression were significantly associated with HBV replication,with higher HBV-DNA-positive HCC than that in HBV-DNA negative group. Conclusion:TGF-β1 and IGF-Ⅱ in HCC tissues are overexpressed and associated with hepatic HBV replication and differentiation degree of HCC.
