摘要
目的:建立高效液相色谱-蒸发光散射检测(HPLC-ELSD)法同时测定柴胡中柴胡皂苷 a、c、d、f 含量。方法:采用Phenomenex C_(18)色谱柱(4.6 mm×250 mm,5μm),以乙腈-水为流动相进行梯度洗脱(0 min:30:70,40 min:50:50,50 min:50:50),流速1.00 mL·min^(-1),Alltech ELSD-2000ES 检测器,漂移管温度:108℃,气体:空气,流速:3.0 L·min^(-1),柱温:室温。结果:柴胡皂苷 a、c、d、f 分别在0.18~8.84,0.17~8.28,0.21~10.40,0.16~8.20μg的范围内线性关系良好(r 分别为0.9997,0.9999,0.9997,0.9997);平均回收率(n=9)分别为103.3%,99.26%,102.0%,101.7%。结论:本法简便、准确,重复性好,可作为柴胡药材质量检测的方法。
Objective:To develop an HPLC -ELSD method for simultaneous determination of saikosaponins a, c, d,f in Radix Bupleuri. Methods:The analysis was performed on a Phenomenex C18 column (4. 6 mm × 250 mm, 5 μm) at the room temperature with a gradient program of acetonitrile -water (0 min:30:70,40 min:50:50,50 min:50: 50) as the mobile phase and an Evaporative Light- Scattering Detector (Alhech ELSD-2000ES) as the detector. The drift tube temperature was 108℃, the gas flow rate was 3.0 L · min^ - 1. Results: Saikosaponins a, c, d,f were linear over the range of 0. 18 - 8.84,0. 17 - 8.28,0. 21 - 10.40,0. 16 - 8.20 μg ( r was 0. 9997, 0. 9999,0. 9997,0. 9997, respectively). The average recoveries ( n = 9 ) were 103.3% , 99. 26% , 102. 0% , 101.7 % for saikosaponins a, c, d, f, respectively. Conclusion: This method is simple, sensitive, reliable and reproducible. It can be used for the quality control of Radix Bupleuri.
