摘要
目的:建立测定质粒 DNA 中乙醇和异丙醇残留量的方法。方法:顶空进样气相色谱法。色谱条件:DB-624石英毛细管柱(30 m×0.53 mm×3μm),柱温:60℃,气化室温度:120℃,FID 检测器,温度为300℃;进样分流比:1:50,载气 N_2(3.0mL·min^(-1))。结果:乙醇、异丙醇线性关系良好(相关系数均大于0.9999);最低检测浓度分别为0.00125 mg·mL^(-1)和0.00083 mg·mL^(-1);平均回收率分别为100.1%和100.9%;平均 RSD 分别为3.0%和1.5%。结论:该法简单快速,可用于质粒 DNA 中乙醇和异丙醇残留量的测定。
Objective:To establish method for rapidly determining ethanol and isopropanol in plasmid. Method: Headspace GC was used. Chromatographic conditions were : capillary column DB - 624 (30m×0.53mm×3μm) was used, column temperature 60 ℃ ; injection temperature 120 ℃ ; FID detector, detector temperature 300 ℃ ; Split injection, split ratio 1 : 50 ; Carrying gas : nitrogen ( 3.0 mL · min^ -1 ). Results: The linearities were good ( r 〉 0. 9999) ;the limit of detection were 0. 00125 mg· mL^-1和 0. 00083 mg· mL^-1 ,respectively;the average recoveries were 100. 1% and 100.9% ;the average RSD were 3.02% and 1.50%, respectively. Conclusion: The method is simple and accurate and suitable for determining ethanol and isopropanol in plasmid.
