摘要
目的:建立测定灰绿黄堇药材中延胡索乙素含量的HPLC方法。方法:用甲醇回流提取灰绿黄堇中延胡索乙素,以Sinochrom ODS-BP(4.6mm×200mm,5μm)为色谱柱;流动相为甲醇-0.1%磷酸(用三乙胺调pH值6.0)(75∶25);检测波长:280nm;流速:1.0mL·min^-1。结果:在此色谱条件下,灰绿黄堇药材延胡索乙素可以得到很好的分离,延胡索乙素在6.7-133.6μg·mL^-1,间呈线性关系(r=0.9998)。回收率为98.2%,RSD为1.51%。结论:本方法简便、结果准确可靠,适合灰绿黄堇中延胡索乙素的含量测定。
Objective: To establish the HPLC method for determining the contents of tetrahydropalmatine in Corydalis adunca Maxim. Method: Tetrahydropalmatine was extracted by methanol reflux method. The separation was performed on Sinochrom ODS-BP (4.6mm × 200mm,5μm ). The mobile phase was methanoh0.1% phosphoric acid ( 75:25 ) ( adjusted to pH 6.0 with triethylamine ) .The UV detector was set at 280nm. Results:The calibration curve of tetrahydropalmatine was linear between 6.7-133.6 μg · mL^-1 with being r=0.9998. The average recovery was 98.2% with RSD 1.51%. Conclusion: The HPLC method for determining tetrahydropalamtine in Corydalis adunca Maxim is simple and reliable.
