摘要
目的探讨肿瘤坏死因子相关凋亡诱导配体(TRAIL)联合顺铂和IFN-α应用对人肝癌细胞生长抑制和诱导凋亡的作用。方法用SRB染色法测定TRAIL、顺铂、IFN-α单独或联合应用对人肝癌细胞SMMC7721的体外增殖抑制作用,并检测各组细胞凋亡率。结果TRAIL、顺铂、IFN-α三者联合作用于SMMC7721细胞后,顺铂与IFN-α联用的体外生存率从70.91%±3.43%降至34.81%±3.10%,细胞凋亡率从12.47%±0.91%升至45.50%±6.25%。结论TRAIL、顺铂、IFN-α三者联用能显著提高肝癌细胞SMMC7721生长抑制和诱导凋亡作用。
OBJECTIVE To investigate the synergistic induction of cytoxicity and apoptosis by the combination of tumor necrosis factor related apoptosis inducing ligand (TRAIL) , cisplatin and IFN - α in hepatoma cells SMMC7721. METHODS SRB assay was used for quantitative evaluation of proliferation of SMMC7721. Flow cytometric analyses were used for the assessment of apoptosis rates after different treatment. RESULTS The combination of the three treatments induced significantly higher apoptosis and cytotoxicity than exposure to TRAIL, cisplatin or IFN - α alone , but the combination of two of the three agents did not show synergistic effect .CONCLUSION The combination of TRAIL, cisplatin and IFN -α could significantly increase cytotoxic and apoptosis effect on SMMC7721.
