摘要
目的建立测定人血浆中盐酸西替利嗪浓度的方法。方法色谱柱为Hypersil BDS C18柱,流动相为乙腈-0.05 mol.L-1磷酸二氢铵溶液(37:70);流速1.5 m.lmin-1;检测波长230 nm;盐酸羟嗪为内标。结果线性范围为0.025~1.500μg.ml-1,回归方程为Y=2.482×10-2X+1.586×10-3(r=0.9986),回收率97.7%,日内和日间RSD分别为5.5%、6.6%。结论所建方法简便、准确、快速,适用于临床上盐酸西替利嗪药物动力学的研究。
OBJECTIVE To establish a method to determine the Cetirizine concentration in human plasma. METHODS The chromagraphy column was the Hypersil BDS C18 column with the mobile phase consisted of acetonitrile -0. 05 mol·L^-1 ammonium dihydrogen phosphate(37 : 70). The UV detector wavelength was set at 230 nm, the fluid speed was 1.5 ml·min^-1 and the hydroxyzine was used as internal standard. RESULTS The calibration curve was linear in the range of 0. 025 - 1. 500 μg·ml^-1 with a regressive equation of Y = 2. 482 × 10^-2X + 1. 586 × 10^-3 ( r = 0. 9986) ; The recovery of cetirizine was 97. 7% ; the relative standard deviations were 5.5% (intra day) and 6. 6% (inter day) respectively. CONCLUSION The method is simple , accurate, rapid, and suitable for determination of Cetirizine in the human plasma and pharmacokinetic studies of Cetirizine.
