深圳地区麻疹病毒流行株血凝素蛋白H基因及核蛋白N基因序列分析 被引量:3

摘要 目的探讨深圳地区目前流行的麻疹病毒的基因型别和特征。方法采用B95a细胞分离培养麻疹病毒,通过RT-PCR方法扩增麻疹病毒核蛋白基因C末端450bp片段和血凝素H基因全序列并测序,其序列与Genebank中国内外流行的麻疹病毒8个基因组的代表毒株的序列进行同源性分析。结果分离到1株麻疹病毒,N基因与H1基因型代表毒株相比同源性为98.4%,H基因与H1基因型代表毒株相比同源性为99.5%。结论深圳市麻疹流行毒株属于H1基因型。 Objective To explore the genetic characteristic and genotype of measles viruses circulating in Shenzhen City . Methods Throat swabs were collected for virus isolation. A 450bp fragment of the C- terminal of the nucleoprotein (N) gene and the full sequence of hemagglutinin(H) gene were amplified by RT- PCR and subjected to sequence analysis by using DNAstar software. Results Only 1 strain of measles virus was isolated from the 15 throat- swabs. N gene homogeneity was 98.4 % and H gene homogeneity was 99:5 % when compared with other wild - type viruses of H1 genotype. Conclusion Measles viruses circulating in Shenzhen belong to the H1 genotype.
