摘要 目的建立HPLC法测定人血浆中头孢拉定的浓度。方法采用甲硝唑为内标,色谱柱为YMCODS-A C18(150*4.6mm),流动相为乙腈:甲醇:pH4.5的醋酸盐缓冲液(6:10:84),流速1.0ml/min,检测波长为260nm的条件下,以HPLC测定人血浆中头孢拉定的浓度。结果头孢拉定血清浓度在0.050~32.100μg.ml-1范围内具有良好线性关系,最低定量限为0.050μg.ml-1,方法回收率为97.5%~100.60%,RSD<5%。结论本法简便、准确、灵敏,适用于头孢拉定血药浓度监测和药代动力学研究。 Objective To develop a method for the determination of cefradine inhuman plasma by using HPLC. Method The internal standard was secnidazole, the column was YMC ODS - A C18 (150 * 4.6 mm), and the mobile phase was mixture of acetonitrile, methyl alcohol, and acetate buffer. The flow rate was 1.0ml/min. The detection wavelength was 260nm. Results The calibration curves were linear when the serum cefradine level was in range of 0. 050- 32. 100μg·ml^-1, the limit of detection was 0. 050μg·ml^-1, the method had recovery rate of 97.59 % - 100.60 %, the RSD of within - day and betweenday was less than 10 96. Conclusion The method is simple, sensitive, accurate and suitable in monitoring the plasma level and clinical pharmacokenitics study of cefalxin in human.
