摘要
目的建立测定人血浆中地西泮、硝西泮、氯硝西泮、三唑仑、艾司唑仑的浓度的胶束电动毛细管色谱(MECC)法。方法采用MECC法,最佳测定条件为:电泳缓冲液为10mmol/L的硼砂缓冲液(pH=7.5),含75mmol/L十二烷基硫酸钠(SDS)及25%甲醇,分离电压为20kV,温度为22℃,检测波长为200nm,采用压力进样,进样时间10秒。苯巴比妥为内标,内标法定量。结果5种药物在其线性范围内线性关系良好,r值均大于0.99,日内和日间RSD均小于7%(n=3)。结论本方法操作简便、准确、灵敏度高,适用于临床人血浆中5种苯二氮艹卓类药物同时定性及定量检测。
Objective To establish a micellar elektrokinetic capillary chromatography (MECC) method for simultaneous determination of 5 benzodiazepines: diazepam, nitrazepam, clonazepam, triazolam and estazolam in human plasma. Methods To found MECC, the running buffer was composed of sodium tetraborate 10 mmol/L (pH = 7.5 ), ~odium dodecyl sulfate (SDS) (75 mmol/L) and methanol (25%) as an organic modifier. The voltage and temperature were 20 kV and 22 ~C. The wavelength was 200 nm. Sample was injected into the capillary by applying pressure for 10.0 s. The concentrations were quantified by internal standard method. Results The linear relation of 5 benzodiazepines were better within their ranges ( r 〉 0.99 ). Intra - day RSD and inter - day RSD were less than 7% ( n = 3). Conclusion The method is simple, accurate and sensible, and is suitable for simultaneous qualitation and quantitation of diazepam, nitrazepam, clonazepam, triazolam and estazolam in human plasma.
