摘要
蛋白质酪氨酸磷酸酯酶1B(PTP1B)在胰岛素信号传导过程中起负调节作用,是治疗糖尿病和肥胖症的潜在靶点。本文按作用位点对近年来研究的PTP1B抑制剂进行综述,分析了N-草酰胺苯甲酸类化合物、水杨酸类化合物、取代苯乙酮及肉桂酸类化合物、氨基磺酸类化合物、拟肽类物质、噻吩及苯并噻吩类化合物、苯并呋喃磺酰胺类化合物和哒嗪类化合物八大类抑制剂的结构及药理活性。
Protein tyrosine phosphatase 1B (PTPIB) is one of the key negative regulators of insulin signal transduction pathway. Therefore, small molecular inhibitors of PTP1B can be promising drug candidates. In this review, we discuss eight categories of small molecular PTP1B-specific inhibitors: oxalylarylaminobenzoic acid derivatives, salicylic acid derivatives, substitute acetophenone derivatives or cinnamaldehyde derivatives, sulfamic acid derivatives, peptidomimetics, monocyclic thiophene benzothiophene derivatives, benzofuransulfonamide derivatives, pyridazine analogues, which were all synthesized recently.
