摘要 目的:考察和比较6种常用多糖除蛋白方法对透明质酸的纯化效果。方法:通过6种常用多糖除蛋白方法,即加热法、盐酸法、三氯乙酸法、Sevage法、氯仿法和等电点沉淀法纯化透明质酸,以蛋白含量、葡萄糖醛酸含量及平均分子量为主要指标,对各种方法的纯化精品进行比较。结果:用等电点沉淀法纯化的精品经鉴定与HA对照品基本相符,平均分子量>106Da,杂蛋白含量<0.05%。结论:采用等电点沉淀法纯化效果最好。 Objective:To investigate and compare the effect of six methods on purifying hyaluronic acid (HA). Methods: Six methods are used to purify HA, including heating,hydrochloric acid,TCA,Sevage,chloroform and isoelectric precipitation. Those methods are compared by taking protein content,glycuronic acid content and average molecular weight as main indexes. Results: The characteristic of purified HA by isoelectric precipitation is basically in accordance with the contrast HA through identifying, The molecular weight of HA is more than 106 Da and the impurity of protein content of the purified HA is less than 0. 05%. Conclusion : The isoelectric precipitation is the best.
