摘要
目的探讨环境条件对猪链球菌2型强毒株05ZYH33电转化效率的影响。方法采用电穿孔的方法对05ZYH33强毒株进行外源DNA的转化,通过调整参数,对影响电转化效率的主要因素进行探索。结果电穿孔法可以成功地转化猪链球菌2型强毒株05ZYH33,转化效率最高可达107/μg质粒DNA。结论猪链球菌2型强毒株05ZYH33高效电转化方法的建立,为深入研究该病原菌的功能基因组学奠定了基础。
To approach the effect of environmental factors on eleetrotransformation efficiency of Streptococcus suis type 2 virulent strain 05ZYH33, electroporation was performed on gene pulser with E. coli-S, suis shuttle vectors, and transformation efficiencies were evaluated by varying biological and electric parameters of electroporation. By optimizing these factors, an efficient eleetrotransformation system for Streptococcus suis 05ZYH33 was established with a high efficiency of 107 transformants per p.g of plasmld DNA. These results provide the foundation for the functional study on genomics for this important pathogen.
