摘要
目的:观察当归芍药散精简方(FBD)及其CO2超临界萃取物(P2)对体外培养的大鼠乳鼠皮质星形胶质细胞缺氧损伤的保护作用。方法:SD大鼠po FBD 125mg/kg或P2 16.3mg/kg后,采集含药脑脊液(CSF-FBD/P2);MTT法测定FBD、P2人工脑脊液(ACSF-FBD/P2)及其含药脑脊液(CSF-FBD/P2)对Na2S2O4诱导星形胶质细胞缺氧损伤的保护作用。结果:1mmol/LNa2S2O4作用1h复氧4h可显著降低星形胶质细胞活力(P〈0.01);(0.1—1)mg/LACSF.FBD和5%-15%CSF-FBD可显著促进缺氧损伤星形胶质细胞活力(P〈0.01);(0.01—1)mg/LACSF.P2和15%CSF-P2可极显著或显著促进缺氧损伤星形胶质细胞活力(P〈0.01—0.05)。结论:FBD及其CO2超临界萃取物对星形胶质细胞缺氧损伤具有保护作用。
Objective:To observe the protective effect of FBD, the simplified formula from Danggui Shaoyao San (DSS), and CO2 supercritical fluid extract( P2 ) on the hypoxic astrocyte anoxic injury. Methods: Sampling cerebrospinal fluid containing FBD/P2 (CSF-FBD/P2) after administration with FBD 125 mg/kg or P2 16,3 mg/kg; MTI' assay was used to measure the viability in the damaged astrocyte induced by Na2 S2 04 Results : 1 h hypoxia and 4 h reoxygen significantly decreased the viability in astmcyte(P 〈 0.01);(0.1 - 1 )mg/L ACSF-FBD and 5% - 15% CSF-FBD can substantially increase the viability of astrocyte induced by Na2S204 (P 〈 0.01) ; (0.01 - 1) mg/L ACSF-P2 and 15% CSF-P2 can substantially increase the viability of astrocyte( P 〈 0.01 - 0.05). Conclusion: FBD and P2 could prevent astrocyte from Na2S204-induced hypoxic damage,which was beneficial to the treatment of cerebral ischemia.
