血小板经mPEG-SPA化学修饰后的效果研究

摘要 目的观察mPEG-SPA化学修饰血小板表面抗原的修饰效果,及修饰对血小板寿命、血小板凋亡的影响。方法用mPEG-SPA对血小板进行化学修饰;流式法检测修饰前后血小板CD42a及PS荧光强度变化;体外保存血小板检测其寿命。结果经mPEG-SPA修饰后,血小板CD42a表达率由修饰前的(13.48±2.92)%降至修饰后的(5.03±1.68)%;血小板死亡率及PS表达率随保存时间延长而升高,但修饰前后差异无统计学意义。结论mPEG-SPA可有效修饰遮蔽血小板表面抗原,并对血小板寿命、血小板凋亡无显著影响。 Objective To evaluate the effect of methoxy-polyethylene glycol succinimidyl propionic acid (mPEG-SPA) on the survival of platelets. Method mPEG-SPA coated platelets were analyzed for the expression of CD42a and externalization of phosphotidyl serine (PS) by flow cytometry. Life span of the platelets was evaluated at 22℃. Result After coating with mPEG-SPA, the expression levels of CD42a was reduced from (13.48 ± 2.92)% to (5.03 ± 1.68)%. The death rate and the expression level of PS in mPEG-SPA coated platelets were similar to the non-coated platelets. Conclusion Coating of platelets with mPEG-SPA has no effect on the life span of platelets.
