摘要
目的:探讨氟伐他汀对骨质疏松大鼠骨折愈合过程中白细胞介素6(IL-6)的影响。方法:卵巢切除后3个月已经出现骨质疏松的SD大鼠72只,于股骨中段制造骨折模型,再随机分为给药组和对照组,每组36只,给药组手术次日给予10mg/(kg·d)氟伐他汀灌胃,疗程为6周,对照组给予安慰剂。利用免疫组化及原位杂交检测骨折愈合过程中给药组和对照组骨折后3、7、14、21、28和42d骨痂中IL-6、IL-6mRNA的表达,并用真彩色图像分析仪作图像分析。结果:给药组和对照组在骨折愈合过程中IL-6及IL-6mRNA基因表达的细胞定位无差别,且表达模式基本相似。术后14d,阳性细胞的阳性程度给药组比对照组强,有显著差异(P<0.05)。结论:氟伐他汀对骨质疏松大鼠骨折愈合过程中IL-6的合成分泌有一定促进作用。
Objective:To explore the effect of fluvastatin on interleukin-6 (IL-6) during fracture healing in rats with osteoporosis. Methods:The rat fracture models of middle part of the femur were established in 72 SD rats with osteoporosis who experienced ovariectomy 3 months ago. The rats were randomly and equally divided into medication group and control group,in which the rats were lavaged with fluvastatin of 10 mg·kg^-1·d^-1 and placebo for 6 days since the next day after fracture,respectively. The expressions of IL-6 protein and mRNA in bony callus were determined by immunohistochemical method and in situ hybridization on days 3,7,14,21,28,and 42 of fracture. The images were analyzed with real-color image analysis system. Results:There were no differences in the cellular localizations of IL-6 protein and mRNA between medication group and control group during fracture healing,and the expressions of IL-6 protein and mRNA were similar in these 2 groups. On day 14 of fracture,the number of cells positive for IL-6 in medication group was higher than that in control group (P 〈 0.05). Conclusion:Fluvastatin can promote the synthesis and secretion of IL-6 during fracture healing in rats with osteoporosis.
