摘要
目的:探讨重组人肝细胞生长因子(rh-HGF)对大鼠心脏移植冠状血管内膜诱生型一氧化氮合酶(iNOS)表达的影响及意义。方法:近交系健康雄性Wistar大鼠和SD大鼠,建立腹腔异位心脏移植模型。实验分3组:同系移植组,异系移植包括对照组(CsA组)和实验组(HGF组)。分别于术后15d、60d采集各组标本,沿冠状动脉切取心肌组织,免疫组化检测冠状血管内膜iNOS的表达,RT-PCR检测冠状血管内膜iNOSmRNA表达,观测冠状血管病理变化。结果:实验组移植心脏冠状动脉内皮细胞完整,内膜轻度增厚,管腔轻度狭窄,病理改变明显轻于对照组。iNOS在移植心脏冠状动脉内膜的表达,实验组均明显高于对照组及同系移植组(P<0.01)。结论:rh-HGF通过上调移植心脏冠状血管内膜iNOS表达,可保护冠状血管内皮功能,预防移植心脏血管病的发生。
Objective:To investigate the effect of human recombinant hepatocyte growth factor (rh-HGF) on the expression of iNOS in the intima of allograft vessel after cardiac transplantation in rats. Methods:The model of heterotopic heart transplantation were established in Wistar and SD rats. The recipient rats were divided into 3 groups (n=20 in each group):isograft group, allograft control group (CsA group),and allograft experiment group (rh-HGF group). The cardiac grafts were harvested 15 and 60 days after transplantation. The expressions of iNOS and iNOS mRNA in the intima of allograft vessel were detected by immunohistochemistrical staining and reverse transcription polymerase chain reaction,respectively. The pathological changes in allograft coronary vessels were observed. Results:In rh-HGF group,the allograft coronary artries showed mild intimal thickening,and the endothelium and internal elastic lamina remained almost intact after transplantation. The expressions of iNOS in the intima of allograft vessels after transplantation in rh-HGF group were significantly up-regulated than those in CsA group and isograft group (P 〈 0.01). Conclusion:rh-HGF may prevent the progression of cardiac allograft vasculopathy by up-regulating the expression of iNOS.
