小鼠单纯疱疹性角膜基质炎复发过程中Tim-3在角膜组织中的表达 被引量:1

摘要 目的:通过检测小鼠单纯疱疹性角膜基质炎(HSK)复发过程中T细胞免疫球蛋白黏蛋白分子3(Tim-3)mRNA在角膜组织的表达水平,探讨Tim-3的表达与HSK复发之间的关系。方法:建立复发性HSK的BALB/c小鼠模型,用紫外线B光照射鼠的角膜诱导HSK复发;分别于紫外线照射前、照射后第3、7、10、14、21及28d,采用RT-PCR方法检测全长的膜型Tim-3(flTim-3)和可溶型Tim-3(sTim-3)在角膜组织中的表达水平;裂隙灯显微镜下检查角膜基质炎程度。结果:处于病毒潜伏感染期的小鼠经紫外线照射诱导角膜基质炎复发后,角膜基质混浊于紫外线照射后的7~14d达到高峰;sTim-3于3~14d高表达,flTim-3于14~28d高表达。结论:sTim-3的高表达与角膜基质炎的复发和发展有关,flTim-3可能有助于角膜基质炎的恢复。 Objective:To investigate the expression of T cell immunoglobulin-and mucin-domain-containing molecule-3 (Tim-3) mRNA in the cornea of mice with recurrent herpetic stromal keratitis (HSK),and to explore the relationship between Tim-3 mRNA and recurrent HSK. Methods: The mouse model of HSK was established in BALB/c mice,and recurrent HSK was induced by ultraviolet-B irradiation. The expressions of full-length membrane-anchored form of Tim-3 (flTim-3) and soluble Tim-3 (sTim-3) mRNA were detected by reverse transcription chain polymerase reaction before and 3,7,10,14,21,and 28 after the irradiation. The degree of keratitis was observed with the slit lamp. Results: After the ultraviolet-B irradiation,the corneal stromal opacification reached the peak on days 7 to 14 in mice in the incubation period,and sTim-3 and flTim-3 were highly expressed on days 3 to 14 and days 14 to 28,respectively. Conclusion: The high expression of sTim-3 in the cornea is correlated with the recurrence and development of HSK,and flTim-3 may contribute to the rehabilitation of HSK.
