灵芝对高糖培养的大鼠肾小球系膜细胞MMP-2和TIMP-2表达的影响

摘要 目的探讨灵芝对在高糖刺激下大鼠肾小球系膜细胞(MCs)表达基质金属蛋白酶-2(MMP-2)及基质金属蛋白酶-2组织抑制剂(TIMP-2)的影响。方法大鼠MCs在高糖培养下,予不同质量浓度灵芝、苯那普利刺激后,运用免疫组织化学技术和图像分析法,观察MCs表达MMP-2和TIMP-2的变化,RT-PCR检测MMP-2、TIMP-2 mRNA表达水平。结果不同质量浓度的灵芝对高糖培养的大鼠MCs MMP-2和TIMP-2表达的影响不同。与高糖组相比,随着灵芝浓度逐渐增加,MMP-2的表达逐渐增加,TIMP-2的表达逐渐降低。结论灵芝通过改变高糖培养的大鼠MCsMMP-2和TIMP-2的表达量,影响糖尿病肾病(DN)的进程。 Objective To investigate the effect of Ganoderma lucidum on the expressions of matrix metalloprotein-ase - 2 ( MMP - 2) and tissue inhibitor of metalloproteinase - 2 ( TIMP - 2 ) by rat glomerular mesangial cells (MCs) cultured under high - glucose environment. Methods The culture of rat MCs was continued under high glucose level for 48 h before the experiment. Following the addition of Ganoderma lucidum of varied concentrations or benazepril, the changes in the expression of MMP - 2 and TIMP - 2 in the MCs and the mRNA levels of them were examined by immunohistochemical method, image analysis and RT - PCR respectively. Results The levels of MMP - 2 and TIMP - 2 were found varying with the concentration of Ganoderma lucidum, i.e. MMP - 2 increased and TIMP - 2 decreased with the increase of Ganoderma lucidum concentration. Conclusion Ganoderma lucidum may modulate the process of diabetic nephropathy by changing the expression of MMP -2 and TIMP - 2.
