参芪复方对GK大鼠2型糖尿病大血管病变CD36 mRNA及ox-LDL的影响 被引量:12

Effects of Shenqi Compound Recipe on CD36 mRNA and ox-LDL in GK Rats with Type Ⅱ Diabetes Mellitus Macrovascular Complications

摘要 目的观察参芪复方对GK大鼠2型糖尿病大血管病变胸主动脉CD36 mRNA表达及血清ox-LDL的影响。方法SPF级GK大鼠(自发性糖尿病大鼠)44只及Wistar大鼠11只按血糖水平随机分为正常对照组、模型组、雷米普利组、参芪复方低剂量组、参芪复方高剂量组。4组GK大鼠组造模:10 mg.kg-1.d-1腹腔注射N-硝基-L-精氨酸甲酯(L-NAME),同时给高脂饮食,连续14 d造模。正常对照组给生理盐水。2周后开始给药,各组分别给相应受试药物28天。用ELISA法测ox-LDL,用实时定量PCR法测胸主动脉CD-36 mRNA表达。结果参芪复方高、低剂量组的血清ox-LDL含量较模型组明显降低(P<0.05),胸主动脉CD36 mRNA的表达显著低于模型组组(P<0.01),血清ox-LDL含量与胸主动脉CD36 mRNA的C t值进行相关性分析,相关系数r=-0.60,P<0.01。结论以益气养阴、清热生津、活血化瘀为治则的参芪复方能明显下调GK大鼠内皮细胞损伤的CD36 mRNA的表达,抑制CD36的生成,可以阻断CD36对ox-LDL的摄取,可能是参芪复方治疗T2DM大血管病变的机制之一。 Objective To explore the effects of Shenqi compound recipe treating type Ⅱdiabetes mellitus (T2DM) on macrovascular complications in GK rats and correlation with thoracic aorta CD36 mRNA and serum Oxidated low density lipoprotein (ox- LDL). Methods GK rats were randomly divided into Model group, Ramipril group, Shenqi compound Recipe low dosage group, Shenqi compound recipe high dosage group and wistar control group. Each group was administrated correspondent substance respectively for 28 days after intra - peritonelly L - NAME 2 weeks except wistar control group. Determined ox - LDL in serum by ELISA. The mRNA expression of CD36 in aorta by real time RT - PCR. Compared the correlation with ox - LDL and CD36 mR- NA. Results Content of ox - LDL decreased in Shenqi compound recipe low dosage and high dosage group compared with model group (P 〈0.05). The expression of thoracic aorta CD36 mRNA in Shenqi compound recipe low dosage and high dosage group also decreased remarkably compared with model group ( P 〈 0. 01 ). Ox - LDL and CD36 mRNAg Ct had correlated negatively with lined( r = -0.60 ,P 〈 0.01 ). Conclusion Shenqi compound recipe with principle tonifying "Qi" and "Yin" and promoting blood circulation to dispate stasis can decreased the production of CD36, inhibited the expression of thoracic aorta CD36 mRNA in GK rats. May be it was one of the mechanism of Shenqi compound recipe treating T2DM on macrovascular complications.
