摘要
目的探索枸杞RAPD反应体系并对其进行优化。方法在本实验室常用的RAPD反应体系的基础上,根据RAPD扩增效果确定枸杞基因组DNA的最佳RAPD反应体系。结果建立了适合枸杞的RAPD反应体系:20μl体系中含1×PCR buffer,50 ng模板,0.4μM 10碱基随机引物、1UTaqDNA聚合酶、2 mM Mg2+,125μMdNTPs。结论枸杞的RAPD反应体系能够用于枸杞的RAPD分析。
Objective To study the RAPD reaction system in Lycium barbarum L. and optimization of it. Methods Based on the RAPD reaction system was often used in our lab, RAPD reaction system was tested by the result of RAPD. Results An RAPD reaction system suitable for Lycium barbarum L. was established, that was, 20μl amplification containing I x PCR buffer,50ng template DNA ,0.4μm primer, 1UTaq DNA polymerase,2 mmol/L Mg^2+ , 125μmol/L dNTPs. Conclusion The RAPD reaction system can be used to RAPD analysis in Lycium barbarum .
