舒肝解郁饮对抑郁大鼠脑组织单胺类神经递质及其代谢产物的影响 被引量:5

摘要 目的:探讨舒肝解郁饮(SGJYY)对抑郁大鼠模型脑组织中单胺类神经递质及其代谢产物的影响。方法:用孤养加慢性不可预见的中等强度刺激建立大鼠抑郁模型,给予中药舒肝解郁饮,用高效液相-电化学方法检测抑郁大鼠脑组织单胺类神经递质及其代谢产物的含量。观察各组大鼠体重和糖水消耗量、旷场行为实验、游泳不动时间等行为学指标的变化。结果:与对照组比较,各造模组大鼠体重增长缓慢(P<0.01),糖水消耗量减少(P<0.01),旷场行为实验得分减少(P<0.01),游泳不动时间延长(P<0.01);与对照组比较,模型组大鼠脑组织5-羟色胺(5-HT)含量降低(P<0.01),5-羟吲哚乙酸(5-HIAA)升高(P<0.01),去甲肾上腺素(NE)降低(P<0.01),5-甲氨基-4羟基苯乙二醇(MHPG)升高(P<0.01);多巴胺(DA)、3,4二羟基苯乙酸(DOPAC)降低(P<0.01)。舒肝解郁饮能对抗慢性应激所造成的大鼠体重增长缓慢,并使各项行为学指标得到改善。百忧解组、SGJYY大剂量组大鼠脑组织中5-HT升至正常水平,SGJYY中剂量组5-HIAA降至接近正常水平;百忧解组、SGJYY大剂量组NE升至正常水平,百忧解组、SGJYY大、中、小剂量组MHPG降至正常水平;百忧解组、SGJYY大剂量组DA升至接近正常水平,百忧解组DOPAC升至接近正常水平。结论:舒肝解郁饮抗抑郁机制与调节脑内单胺类神经递质5-HT、NE、DA及其代谢物的含量有关。 Objective To study the effect of Shuganjieyuyin (SGJYY) on monoamine transmitters and their metabolites of brain tissue in depressive mice. Methods The lonely feeding and chronic moderate intensive unpredictable stimulation were used to build depression model and SGJYY was administered. HPLC was used to test the contents of monoamine transmitters and their metabolites of brain tissue in depressive mice and observe the changes of weight and consumption of sugar water and behaivor indexes in mice. Results Compared with normal group, the weight increased slowly (P〈0.01), the score of Open-field and sugar water consumption decreased (P〈0.01), the time of no-moving increased (P〈0.01) in model group. Compared with normal group, the contents of 5-HT, NE, DA and DOPAC of brain tissue decreased (P〈0.01), 5-HIAA and MHPG increased (P〈0.01) in model group. The 5-HT contents in BYJ and large dosage SGJYY groups were near normal level, the 5-HIAA content in middle dosage SGJYY group was near normal level. The contents of NE in BYY and large dosage SGJYY groups were near normal level, the contents of MHPG in BYJ and anti-depression of SGJYY groups were near normal level. The contents of DA in BYJ and large SGJYY group were near normal level, the DOPAC content in BYJ group was near normal level. Conclusion The mechanism of anti-depression of SGJYY is related to regulating the contents of monoamine transmitters and their metabolites.
