摘要
目的探讨雌激素对雌激素受体阳性人乳腺癌细胞株MCF-7和B37组织激肽释放酶5(KLK 5)mRNA的表达水平,以及雌激素对KLK5 mRNA表达水平的影响。方法分别取生长良好的MCF-7和B37细胞,在不同浓度的雌激素(17-βE2)存在下培养72小时后,收集细胞,提取总RNA,采用荧光定量逆转录-聚合酶链反应,检测KLK5 mRNA表达水平及变化。结果在未经刺激的乙醇对照组,MCF-7细胞KLK5 mRNA的相对表达水平(KLK5 mRNA/GAPDH mR-NA)为(3.23±0.51)×10-5,B37细胞KLK5 mRNA表达水平为(3.13±0.62)×10-4。B37细胞KLK5 mRNA表达水平高于MCF-7细胞。当17-βE2浓度10-11mol/L以下时,MCF-7和B37细胞中KLK5 mRNA表达水平与乙醇对照组无明显差异(P<0.05)。随17-βE2浓度的增高,MCF-7和B37细胞中KLK5 mRNA的表达呈递增趋势,各组与乙醇对照组相比差异有显著意义(P<0.01)。结论雌激素对人乳腺癌细胞株MCF-7和B37细胞的KLK5 mRNA表达有剂量依赖的上调作用。
Objective To investigate the effects of estrogen on the expression of KLX5 mRNA in human breast cancer cell line MCF-7 and B37. Methods MCF-7 and B37 cells were incubated with 17-βE2 at different concentration for 72 hours, respectively. The expression levels of KLK5 mRNA were evaluated by fluorescence quantitative RT-PCR.Results KLX5 mRNA expression levels (KLX5 mRNA/GAPDH mRNA)in MCF-7 and B37 were (3.23 ± 0.51 ) × 10^-5 and (3.13 ± 0.62) × 10^-4, respectively. Compared with ethanol control, KLK5 mRNA expression levels in MCF-7 and B37 were significantly increased with the increase of 17-βE2 concentration ( P 〈 0.01 ), while showed no statistical change when 17-βE2 concentration was below 10^-11 mol/L( P 〈 0.05). Conclusion Estrogen up-regulated the expression levels of KLX5 mRNA in MCF-7 and B37 in a dose dependent way.
