摘要
目的比较海洛因对大鼠C6胶质瘤细胞的双向电泳图谱差异,从蛋白质水平初步探索阿片类毒物-海洛因的作用机制。方法提取对照组与海洛因组C6胶质瘤细胞的蛋白质,以固相pH梯度等电聚焦为第一向,SDS-PAGE垂直电泳为第二向进行2-DE。以图像分析软件ImageMaster V 5.0分析电泳图谱。结果将对照组与海洛因组C6胶质瘤细胞的2-DE图谱匹配后,检测到570个蛋白点,差异大于1.5倍的斑点有29个,其中20个下调,9个上调。结论初步建立了大鼠C6胶质瘤细胞的蛋白质组学分析方法;差异点的发现为深入理解海洛因的分子机制提供了线索。
Objective To investigate the molecular mechanisms of heroin by comparing two-dimensional electrophoregram of the C6 glioma cell with heroin and sodium chloride. Methods The C6 glioma cell proteins was extracted. The electrophoregrarns were obtained with immobilized pH gradient(IPG) isoelectric focusing electrophoresis as the first dimension, and then with vertical SDS-PAGE as the second. The electrophoregrams were visualized using silver staining and analyzed with ImageMaster V 5.0 software. Results 570 protein spots could be matched, 29 protein spots with significant difference were found,20 protein spots were decreased,while 9 protein spots were increased. Conclusion Proteomics method of C6 glioma cell was established. The differentially displayed proteins may provide further insight into molecular mechanisms of heroin.
