摘要 目的探索无血清培养系统用于结膜上皮杯状细胞培养的可行性。方法用不含血清培养液进行组织块培养,通过倒置相差显微镜观察细胞形态,并应用特殊组化染色和免疫组化方法对培养杯状细胞进行鉴定。结果倒置相差显微镜下可见在无血清培养液中杯状细胞形态与活体组织中相似,免疫组化法证实部分细胞PAS和MUC5AC呈阳性染色。结论兔结膜杯状细胞可通过无血清培养法在体外培养,并保持活体细胞特性,为进一步研究杯状细胞相关的眼表疾病及药理毒理作用提供有效的细胞模型。 Objective To investigate the facibihty of serum-free culture system on culturing eonjunetival goblet cells. Methods Rabbit conjunctival epithelial tissues were explanted in serum-free system. The goblet cells were identified with inverted phase contrast microscope, histochemistry and immunohistochemistry. Results Goblet cells were successfully cultured in serum-free medium. These ceils not only demonstrated a normal structural, but also retain the PAS and MucSAC positive staining. Conclusion Rabbit goblet cells that retain characteristics of goblet cells in vivo could be cultured in a serum-free culture system. This may serve as a useful tool in the basic research and the research of ocular surface diseases related to goblet cells.
